So she took a fit, said she wasn’t happy, and crap like that about our relationship, said she hasn’t been happy the past few weeks….
Well, I don’t think it’s fair because when i try to talk to her sometimes she’ll completely ignore me, then get upset because I persist on talking to her?
I love her alot, but I don’t understand why she keeps telling me that she wants to end the relationship? Even yesterday she said, it’s not you it’s me bull crap….

The paleoherb species Asarum caudigerum (Aristolochiaceae) is important for research into the origin and evolution of angiosperm flowers due to its basal position in the angiosperm phylogeny. In this study, four MADS-box-containing transcripts were isolated from A. caudigerum by rapid amplification of cDNA ends (RACE). Sequence comparisons and phylogenetic analyses indicated that they possess high homology to AP3 subfamily genes, which have been shown previously to be involved in petal and stamen development in eudicots. Reverse-transcription quantitative PCR (RT-qPCR) and in situ hybridization analyses showed AcAP3-A expression mainly in the second whorl (stamens) and AcAP3-B expression in whorls 1 and 3 (perianth and carpels). Compared with eudicot AP3 homologs, premature translation termination codons were caused by an insertion in the K1 domain of AcAP3-C, and by a deletion in the 7th exon of AcAP3-D. Sequence analyses suggested that the A. caudigerum AP3 lineage had undergone gene duplication and subfunctionalization, diverging in expression patterns during perianth, stamen, and carpel development. Based on comparative genomic and phylogenetic analyses, we concluded that subfunctionalization has likely contributed to the persistence of two functional AP3 paralogs, that two other copies may have become pseudogenes, and that these AP3 duplication and subfunctionalization events may have contributed to the evolution of the unusual floral morphology of A. caudigerum.

Nectarivorous birds and bats have evolved highly specialized tongues to gather nectar from flowers. Here, we show that a nectar-feeding bat, Glossophaga soricina, uses dynamic erectile papillae to collect nectar. In G. soricina, the tip of the tongue is covered with long filamentous papillae and resembles a brush or mop. During nectar feeding, blood vessels within the tongue tip become engorged with blood and the papillae become erect. Tumescence and papilla erection persist throughout tongue retraction, and nectar, trapped between the rows of erect papillae, is carried into the mouth. The tongue tip does not increase in overall volume as it elongates, suggesting that muscle contraction against the tongue’s fixed volume (i.e., a muscular hydrostat) is primarily responsible for tip elongation, whereas papilla erection is a hydraulic process driven by blood flow. The hydraulic system is embedded within the muscular hydrostat, and, thus, intrinsic muscle contraction may simultaneously increase the length of the tongue and displace blood into the tip. The tongue of G. soricina, together with the tongues of nectar-feeding bees and hummingbirds, which also have dynamic surfaces, could serve as valuable models for developing miniature surgical robots that are both protrusible and have highly dynamic surface configurations.

hi me and my girlfriend and daughter who is 4 live in a housing association house .
its is a lovely house in a nice area , our neighbours both work like us and a few others and we keep our houses nice and gardens nice.
my neighbours also keep the entrance to where we live nice by planting flowers and put pots out there so it looks nice ,its in the car park area,
the thing is some of the residents the ones that don’t work have been saying my neighbours are posh and taking over because there making it look nice ,
I cant believe they said that and said to them that they only want it to look nice and they are doing no harm they are a decent family what’s wrong with you would you rather have rough people in here,
what do you think do you see anything wrong with what my neighbours are doing.

BACKGROUND: Non-viral lipid-based gene delivery vectors have been shown to possess better stability and longer circulation time after surface PEG modification. However, surface PEGylation may decrease the transfection efficiency dramatically. Here, we address the hypothesis that down regulating the lysosomal processing with a clinical available proton pump inhibitor omeprazole might decrease sequestration of PEGylated lipid-mu-DNA (LMD) in intracellular organelles, thereby increasing their transfection efficiencies. METHODS: LMD nanoparticles were prepared by the self-assembling of cationic liposomes, peptide mu and plasmid DNA. The characteristics of LMD lipopolyplexes were detected by scanning electron microscopy (TEM), photon correlation spectroscopy (PCS) and DNA gel electrophoresis. They were added into cultured cells with or without omeprazole pretreatment. The detailed cellular uptake and subcellular distribution were followed by flow cytometry and confocal microscopy. The gene transfection efficiencies were evaluated in four cell lines as well as in xenograft tumor models. RESULTS: Lysosome staining revealed that 0.1 mg/ml (290 μM) omeprazole raised the pH value of intracellular acidic organelles and induced alterations in the lysosomal compartments. Confocal microscopy showed that more gene materials distributed from intracellular organelles to cytoplasms with omeprazole treatment. Luciferase gene transfection assay showed that omeprazole at probably non-toxic concentration increased PEGylated LMD transfection efficiency significantly in human (NCI-H1299, HT-1080 and A375) and mouse (4 T1) cell lines, as well as in tumors of H1299 xenograft tumor models (p
